Materials and Equipment:

Tranexamic acid sample
Sodium 1,2-naphthoquinone-4-sulfonate (CAS number: 521-24-4)
Sodium hydroxide (NaOH) solution (0.1 M)
Deionized water
UV-VIS spectrophotometer
Quartz cuvettes
Volumetric flasks and pipettes
Spectrophotometer software (for data analysis)


Procedure:

Preparation of Reagents:

a. Prepare a stock solution of Sodium 1,2-naphthoquinone-4-sulfonate by dissolving an appropriate amount of the reagent in deionized water. The exact concentration will depend on specific experimental conditions.

b. Prepare a 0.1 M sodium hydroxide (NaOH) solution.

Sample Preparation:

a. Weigh a known amount of tranexamic acid sample accurately using an analytical balance. Record the mass (in grams) of the sample.

b. Dissolve the tranexamic acid sample in deionized water to prepare a solution. The concentration should be within the linear range of calibration curve.

Reaction and Color Development:

a. Transfer a measured volume of the tranexamic acid solution to a suitable container.

b. Add a measured volume of the Sodium 1,2-naphthoquinone-4-sulfonate solution to the container. The reaction between tranexamic acid and the reagent will result in the formation of a colored complex.

c. Allow the reaction to proceed for a specific period of time, usually around 10-15 minutes, at room temperature or at a specified temperature.

UV-VIS Spectrophotometer Setup:

a. Turn on the UV-VIS spectrophotometer and allow it to warm up for the specified time.

b. Set the spectrophotometer to the appropriate wavelength for measuring the absorbance of the colored complex. The choice of wavelength will depend on the specific reaction product formed.

c. Ensure the spectrophotometer is blanked with a cuvette containing the reagent solution without the tranexamic acid sample.

Measurement:

a. Fill a quartz cuvette with the reaction mixture containing the colored complex.

b. Place the cuvette into the sample holder of the UV-VIS spectrophotometer.

c. Record the absorbance of the colored complex at the chosen wavelength.

Data Analysis:

a. Create a calibration curve by plotting the absorbance values against known concentrations of tranexamic acid.

b. Use the calibration curve to determine the concentration of tranexamic acid in unknown sample by measuring its absorbance at the same wavelength.
