    Bifida Repair Complex™ (Bifida Ferment Lysate, High Conc) 5%

    Bifida Repair Complex™, a highly concentrated fermentation extract from Bifidobacterium, on enhancing skin barrier function and resisting damage from oxidative stress and inflammation using in vitro models.



    Skin barrier enhancement:


    - BFL upregulated the expression of genes involved in the skin physical barrier (filaggrin, loricrin, involucrin, transglutaminase 1, aquaporin 3) and antimicrobial barrier (cathelicidin, β-defensin 2) in HaCaT keratinocytes in a dose-dependent manner.
    - BFL treatment increased filaggrin protein levels in HaCaT cells.
    - These results suggest BFL can promote skin barrier maintenance by inducing expression of barrier-related molecules.



    Antioxidant effects:


    - BFL exhibited strong free radical scavenging capacity against DPPH, ABTS, hydroxyl, and superoxide radicals in a dose-dependent manner, with highest activity against hydroxyl radicals.
    - In H2O2-induced oxidative stress in HaCaT cells, BFL treatment dose-dependently:
        - Increased activities of antioxidant enzymes catalase and glutathione peroxidase
        - Reduced intracellular reactive oxygen species and malondialdehyde levels
    - Indicating BFL can protect cells against oxidative damage by boosting antioxidant capacity.



    Anti-inflammatory effects:


    - In lipopolysaccharide (LPS)-stimulated THP-1 macrophages, BFL significantly inhibited secretion of inflammatory cytokines IL-8 and TNF-α.  
    - BFL decreased expression of the inflammatory mediator cyclooxygenase-2 (COX-2).
    - Suggesting BFL can modulate the immune response and reduce inflammation.



    In summary, the results showed that BFL can reinforce the skin barrier by upregulating barrier genes/proteins and antimicrobial peptides, while also protecting against oxidative stress through antioxidant activity and reducing inflammation by modulating cytokine secretion. The authors conclude BFL strengthens skin defenses against environmental insults like oxidation and inflammation.
