  1. Inhibition of MMP-1 Expression: Rg2 was shown to downregulate the expression of matrix metalloproteinase-1 (MMP-1) in human fibroblasts (Hs68 cells) that were treated with UVB-irradiated keratinocyte-conditioned medium (UV CM). MMP-1 is a collagenase that plays a key role in collagen degradation, which is associated with skin aging and wrinkle formation.

  2. Concentration-Dependent Effects: The inhibitory effect of Rg2 on MMP-1 gene and protein expression was found to be concentration-dependent. Specifically:

    • At 12.5 µM, 25 µM, and 50 µM concentrations, Rg2 significantly inhibited MMP-1 expression without causing cytotoxicity in Hs68 cells.
    • The maximum effect was observed at a concentration of 50 µM.
  3. Mechanism of Action: The study showed that Rg2 inhibits the phosphorylation of extracellular signal-regulated kinase (ERK) and c-Jun N-terminal kinase (JNK), which are part of the mitogen-activated protein kinase (MAPK) signaling pathway. However, Rg2 did not affect the phosphorylation of p38 MAPK. This suggests that Rg2 interferes with specific pathways that lead to MMP-1 expression.

  4. Cytotoxicity Assessment: Rg2 at concentrations up to 50 µM did not exhibit cytotoxic effects on Hs68 cells. However, a concentration of 100 µM combined with UV CM showed cytotoxicity, reducing cell viability to 78.9 ± 2.55%.

  5. Implications for Photoaging Prevention: The ability of Rg2 to inhibit MMP-1 suggests it has potential applications in preventing photoaging, which is the aging of the skin caused by exposure to ultraviolet (UV) radiation.
